Colchicine inhibits microtubule polymerization (IC50 = 3.2 uM) by binding to tubulin, which disrupts spindle formation during mitosis. Since increased rate of mitosis is associated with cancer cell proliferation, blocking microtubule function with colchicine has been used as an approach to anti-cancer therapy. For instance, colchicine blocks the growth of MCF-7 human breast carcinoma cells with an IC50 value of 13 nM. Its mitosis-inhibiting function is also utilized to perform karyotypes in genetic studies. Colchicine produces anti-inflammatory effects by inhibiting neutrophil motility and activity (5 umol/kg in a mouse model of gouty arthritis) and inhibits the deposition of uric acid, a key aspect in the treatment of gout.
