- The unusual amino acid hypusine is formednposttranslationally and is only found in a single cellular protein,neukaryotic translation initiation factor 5A. In the first step ofnhypusine biosynthesis, deoxyhypusine synthase catalyzes thenNAD-dependent transfer of the butylamine moiety of spermidine tonthe epsilon-amino group of a specific lysine residue of the EIF5Anprecursor protein to form the intermediate deoxyhypusine residue.nThis gene consists of nine exons spanning 6.6 kb. Three transcriptnvariants have been isolated. However, only transcript variant 1nencodes an active protein. The shorter variants may act asnmodulating factors of DHPS activity.
