ω-conotoxins are small, disulfide-rich peptides from the venoms of predatory cone snails that inhibit neuronal voltage-gated Ca2+ channels involved in neurotransmitter release. ω-conotoxin-MVIID (ω-CTx-MVIID) was identified by sequence homology from Conus magus (Magician's cone snail) cDNA library. ω-CTx-MVIID blocks Ca2+ current activated in whole-cell patch-clamp recordings from central neurons of goldfish. ω-conotoxin MVIID exhibits greater discrimination against the N-channel subtype than any other ω-conotoxin variant to date. ω-conotoxin MVIID has effects on “P-like” subtypes of Ca2+ channels. Furthermore, ω-CTx-MVIID inhibits synaptosomal voltage-sensitive Ca2+ uptake.
