IL-8 (CXCL8) is a small cytokine belonging to the chemokine CXC subfamily. There have been 17 different CXC chemokines described in mammals. The members of the CXC chemokine subfamily have been subdivided into two categories - those with a specific amino acid sequence of glutamic acid-leucine-arginine (ELR) immediately before the first cysteine of the CXC motif (ELR-positive), and those without an ELR motif (ELR-negative). ELR-positive CXC chemokines specifically induce the migration of neutrophils, and interact with chemokine receptors CXCR1 and CXCR2. IL-8 is an ELR-positive CXC chemokine. The primary function of IL-8 is to recruit neutrophils to the site of inflammation. It also activates neutrophil functions and promotes angiogenesis. Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.
