C反应蛋白(CRP)是肝脏对炎症反应产生的一种物质。它是身体炎症的标志物,经常在医疗环境中用于评估炎症的存在和严重程度。CRP水平会因各种情况而升高,包括感染、损伤和慢性炎症性疾病。
血液中高水平的CRP可以表明炎症的存在,但不能确定确切的原因。它通常与其他临床和实验室发现结合使用,以帮助诊断和监测感染、自身免疫性疾病和心血管疾病等疾病。CRP水平升高也可能与类风湿性关节炎、炎症性肠病和某些癌症等疾病有关。
在双抗体夹心狗/犬CRP ELISA测定中,样品中存在的CRP与已吸附到聚苯乙烯微孔表面的抗狗CRP抗体反应。在通过洗涤去除未结合的蛋白质后,加入与辣根过氧化物酶(HRP)缀合的抗狗CRP抗体。这些酶标记的抗体与先前结合的CRP形成复合物。在另一个洗涤步骤之后,通过添加显色底物3,3’,5,5’-四甲基联苯胺(TMB)来测定与免疫吸附结合的酶。结合酶的量直接随测试样品中犬CRP的浓度而变化;因此,在450nm处的吸光度是测试样品中CRP浓度的测量值。试验样品中CRP的量可以根据标准曲线进行插值,并根据样品稀释度进行校正
