DV4-E88单克隆抗体与登革热病毒血清型4(DENV-4)E蛋白上的DIII结构域反应。这种血清型特异性抗体不会与登革病毒1型、登革病毒2型或登革病毒3型(即登革病毒的其他血清型)发生交叉反应。登革病毒是一种由埃及伊蚊和白纹伊蚊传播的黄病毒,每年在热带和亚热带地区造成超过1亿人感染。轻度登革热的特征是发烧、头痛和肌痛,而重度登革热可导致出血和毛细血管渗漏综合征,即登革热出血热(DHF)/登革热休克综合征(DSS)。从结构上讲,DENV是一种单链正极性RNA病毒,其包膜(E)蛋白有三个结构域。DV4-E88单克隆抗体(构象地)结合DENV-4(E二聚体上的四表位)上结构域III(DIII)的侧脊(LR)。该抗体不识别酵母表面表达或分离的DENV-4 E蛋白DIII。DIII-LR区含有宿主细胞表面受体识别位点,参与病毒中和抗体的诱导。在体外研究中,DV4-E88单克隆抗体中和了1036株、TVP-376株和TVP-986株DENV-4基因型II毒株。在涉及AG129小鼠的实验中,在病毒感染前单次体内注射DV4-E88单克隆抗体(100-500?g/小鼠)对DENV-4毒株H-241和TVP-376提供了适度到显著的保护。
