检测原理：

双抗体夹心ELISA的原理如图1所示。在该测定中，样品中存在的IgA与已吸附在聚苯乙烯微量滴定池表面的抗IgA抗体反应。在通过洗涤去除未结合的蛋白质后，加入与辣根过氧化物酶（HRP）缀合的抗IgA抗体。这些酶标记的抗体与先前结合的IgA形成复合物。在另一个洗涤步骤之后，通过添加无色底物3，3’，5，5’-四甲基联苯胺（TMB）来测定与免疫吸附结合的酶。结合酶的数量与所测试样品中IgA的浓度直接相关；因此，在450nm处的吸光度是测试样品中IgA浓度的测量值。试验样品中IgA的量可以根据标准曲线进行插值，并根据样品稀释度进行校正。
