犬尿氨酸是一种非蛋白质性氨基酸,在色氨酸降解过程中作为代谢中间体产生[1-5]。色氨酸的降解是由诱导型酶吲哚胺-2,3-双加氧酶(IDO)催化的。产品为犬尿氨酸[4,6-8]。细胞因子,特别是干扰素γ[5,9,10],会影响IDO的活性,因此犬尿氨酸途径与免疫系统密切相关[9,11]。犬尿氨酸可以进一步转化为具有神经保护作用的犬尿氨酸酯,也可以转化为具有神经毒性的喹啉酸[6,11]。

LDN Labor Diagnostika Nord-犬尿氨酸ELISA预期用途和原理:
酶免疫测定法用于定量测定血清和EDTA血浆样品中的L-犬尿蛋白,以评估L-犬尿素稳态。在酰化过程中,犬尿蛋白在37°C下被激活,随后与蛋白质偶联。随后的竞争ELISA使用微量滴定板形式。抗原与微量滴定板的固相结合。酰化标准品、对照品和样品的分析物浓度与固定数量的抗体结合位点的固相结合分析物浓度相匹配。在系统处于平衡状态后,通过洗涤除去游离抗原和游离抗原-抗体复合物。通过偶联TMB作为底物的抗兔IgG过氧化物酶检测与固相结合的抗体,从而产生显色反应。在450nm的波长下监测反应。通过将未知样品的吸收与用已知标准浓度制备的参考曲线进行比较来实现未知样品的定量。建议手动处理ELISA。自动实验室设备的使用由用户负责。本产品不用于临床诊断。
