单抗分类鉴定首选

在单克隆抗体的制备过程中,杂交瘤细胞的筛选和鉴别是最重要的步骤。首先需要筛选出正确融合的杂交瘤细胞,再筛选出能分泌目标抗体的杂交瘤细胞,最终将筛选出来的杂交瘤细胞进行扩大培养,制备大量单克隆抗体。在进行杂交瘤细胞的筛选时,需要鉴定这类杂交瘤细胞培养上清液中单克隆抗体的类别和亚型,最常用的是ELISA法,因其快速、简便、灵敏度高、特异性强、易于标准化等优点,已得到广泛应用。


单克隆抗体分型ELISA试剂盒应用

单克隆抗体分型ELISA试剂盒运用双抗体夹心法原理,先用Ig捕获二抗在96孔板上包被固相,然后加入待测的杂交瘤细胞培养上清液,其中的目标蛋白便会与96孔板上的固相二抗结合,洗板去除游离物质后加入用于筛选杂交瘤细胞的标记二抗孵育,洗板去除游离物质,加入显色底物显色,在450nm处测定OD值,进行定性及定量分析。


SBA单克隆抗体分型试剂盒

SouthernBiotech单克隆抗体分型试剂盒专为小鼠单克隆抗体的分型设计。分型试剂盒包含纯化的抗小鼠Ig捕获抗体和HRP或AP偶联的抗小鼠Ig、IgA、IgG1、IgG2a或IgG2c、IgG2b、IgG3、IgM、kappa和lambda抗体,用于检测。这些也可用于血清、上清和腹水等样品中小鼠免疫球蛋白水平的定量研究。

试剂盒有多种形式,可用于检测表达Igh1-a等位基因(如BALB/c)和Igh1-b等位基因(如C57BL/6、C57BL/10、SJL、NOD)的小鼠的免疫球蛋白。使用我们的小鼠免疫球蛋白面板与我们的单克隆抗体分型试剂盒可以生成可靠的标准曲线。

 

SBA克隆分型系统- HRP试剂盒(货号5300-05)含有2.5 mg捕获抗体和1.0 mL HRP偶联抗小鼠Ig、小鼠IgA、小鼠IgG1、小鼠IgG2a、小鼠IgG2b、小鼠IgG3、小鼠IgM、小鼠κ、小鼠λ和ABTS底物。

SBA单克隆抗体分型试剂盒优势

可用于初筛抗体高分泌杂交瘤细胞株,鉴定阳性克隆细胞的特异性抗体分型(IgA、IgM、IgG1、IgG2a、IgG2b、IgG3), 鉴定抗体kappa、lambda轻链异构体以及普通抗体分型ELISA研究;

其中5300-01套装内含有小鼠源IgA、IgM、IgG1、IgG2a、IgG2b、IgG3免疫球蛋白标准品,每种均为1ml(浓度为100μg/ml);而5300-01B套装内用IgG2b替换了5300-01里的IgG2c。而每种SBA单克隆抗体分型试剂盒则包含了1ml亲和纯化山羊抗小鼠Ig捕获二抗(用于包被96孔板),1ml酶标记山羊抗小鼠Ig二抗(用于初筛杂交瘤细胞),1ml×8酶标记山羊抗小鼠IgA, IgM, IgG1, IgG2a, IgG2b, IgG3, Ig kappa,Ig lambda二抗,酶反应底物及操作手册;此外还有配套Ig标准品可供选择。
